Sperm viability vitality should be assessed if a low percentage of sperm are progressively motile, eg. Since motile cells are inherently viable, a viability assessment may not be necessary when motility is high. This test is important to determine if the non-motile spermatozoa are alive or dead.
The fundamental test for male infertility, semen analysis, is mostly a manually performed subjective and time-consuming process and the use of automated systems has been cost prohibitive. We have previously developed an inexpensive smartphone-based system for at-home male infertility screening through automatic and rapid measurement of sperm concentration and motility. Here, we assessed the feasibility of using a similar smartphone-based system for laboratory use in measuring: a Hyaluronan Binding Assay HBA score, a quantitative score describing the sperm maturity and fertilization potential in a semen sample, b sperm viability, which assesses sperm membrane integrity, and c sperm DNA fragmentation that assesses the degree of DNA damage.
Sperm motility describes the ability of sperm to move properly through the female reproductive tract internal fertilization or through water external fertilization to reach the egg. Sperm motility can also be thought of as the qualitywhich is a factor in successful conception ; sperm that do not "swim" properly will not reach the egg in order to fertilize it. Sperm motility in mammals also facilitates the passage of the sperm through the cumulus oophorus a layer of cells and the zona pellucida a layer of extracellular matrixwhich surround the mammalian oocyte.
In humans, sperm DNA fragmentation rates have been correlated with sperm viability rates. Reduced sperm viability is associated with high sperm DNA fragmentation, while conversely high sperm viability is associated with low rates of sperm DNA fragmentation. We postulated that in some men, the sperm viability assay could predict the sperm DNA fragmentation rates. This in turn could reduce the need for sperm DNA fragmentation assay testing, simplifying the infertility investigation and saving money for infertile couples.
